Duloxetine inhibits neuronal serotonin and norepinephrine reuptake. Although the specific mechanism of action of the drug’s central pain-inhibitory effect is unknown, it is thought to be connected to the potentiation of serotonergic and noradrenergic activity in the CNS. Blocking norepinephrine reuptake, in particular, has been shown to help with neuropathic pain.
